    • Abstract:
      This research was aimed to evaluate the Solanum ferox/SF and bitter ginger (Zingiber zerumbet/ZZ) in increasing the effectiveness of the Pseudomonas sp. (EP-02) vaccine through immersion in tilapia (Oreochromis niloticus). The dose used was 600 mg.L-1 of SF; 200 mg.L-1 of ZZ and vaccine bacterial density used 108 CFU.mL-1, the ratio between vaccine and extract was 1: 1: 1. The test were begun with immersion the tilapia in the vaccine and extract for 20 minutes, the challenges test was done on day 7th (d7), 14th (d14) and 21th (d21) post-vaccination with the combined bacteria Aeromonas hydrophila (EA-01) and Pseudomonas fluorescens (EP-01) (concentration of each bacteria was 105 CFU.mL-1) through intramuscular injection of 0.1 mL each fish. Abnormal swimming patterns, pathology of anatomy, total mortality, survival rate, RPS, were measured at week 2 after the challenge test. Application of SF and ZZ extracts in vaccination can increase the non specific (haematology parameters) and specific immunity (phagocytic index and antibody) since d7 of vaccination. While the increasing fish immunity in vaccine groups occurred in d14 post vaccination. The extract administration in vaccination reduced the fish mortality; increased SR and RPS of tilapia faster than the vaccination group. In conclusion, addition of SF and ZZ extracts can increase the efficacy of vaccines faster and more effectively from day 7th of infection. [ABSTRACT FROM AUTHOR]

      Penelitian ini bertujuan untuk menganalisis kemampuan ekstrak terung asam (Solanum ferox/SF) dan lempuyang (Zingiber zerumbet/ZZ) dalam meningkatkan efikasi vaksin bakteri Pseudomonas sp. (EP- 02) yang diberikan melalui perendaman pada ikan nila (Oreochromis niloticus). Dosis yang digunakan yaitu ekstrak SF 600 mg.L-1; ZZ 200 mg.L-1 dan kepadatan bakteri vaksin yang digunakan 108 CFU.mL-1, rasio antara ekstrak terung asam, ekstrak lempuyang dan vaksin adalah 1:1:1. Pengujian diawali dengan merendam selama 20 menit ikan nila (O. niloticus) dengan campuran vaksin dan ekstrak tanaman terung asam/SF dan lempuyang/ZZ, dilanjutkan dengan uji tantang dengan bakteri gabungan Aeromonas hydrophila (EA-01) dan Pseudomonas fluorescens (EP-02) (kepadatan bakteri masing-masing 105 CFU.mL- 1) melalui injeksi intramuskular sebanyak 0,1 mL pada hari ke-7 (d7), 14 (d14) dan 21 (d21) pascavaksinasi. Pengamatan imunitas non spesifik dan spesifik serta tingkat kelangsungan hidup diukur pada minggu ke-2 pasca uji tantang. Hasil penelitian menunjukkan bahwa aplikasi ekstrak SF dan ZZ pada vaksinasi mampu meningkatkan imunitas non spesifik (parameter darah) dan imunitas spesifik (indeks fagositik/ IP dan titer antibodi) sejak hari ke-7/d7 pemberian, sedangkan pemberian vaksin saja tanpa ekstrak baru terjadi peningkatan secara signifikan pada hari ke-14/d14 pascavaksinasi. Peningkatan kelangsungan hidup pascainfeksi dengan bakteri A. hydrophila dan P. fluorescens juga lebih tinggi pada ikan yang diberikan vaksin dan ekstrak. Sebagai kesimpulan, penambahan ekstrak SF dan ZZ mampu meningkatkan kinerja vaksin lebih cepat dan lebih efektif sejak hari ke-7/d7 infeksi. [ABSTRACT FROM AUTHOR]
